Our work revealed the missing phosphatase (Protein Phosphatase 1-Bud14) in the Spindle Position Checkpoint (SPOC).We propose a checkpoint model in which two independent mechanisms activate the checkpoint effector Bfa1:
1. Kin4 kinase phosphorylates Bfa1 to rapidly remove Bfa1 away from yeast centrosomes where inhibitory phosphorylation of Bfa1 by the polo like kinase Cdc5 takes place.
2. PP1-Bud14 dephosphorylates Cdc5-phosphorylated Bfa1 to activate the Bfa1-Bub2 GAP complex.
Thus, checkpoint is robustly activated by: (1) isolating the substrate protein from the kinase (2) making the substrate accessible for the phosphatase at the same time.
SPOC is an essential checkpoint for maintenance of ploidy and genetic stability in the budding yeast. A similar checkpoint exists in stem cell divisions of Drosophila (Centrosome Orientation Checkpoint) and may be yet to be discovered in many other asymmetrically dividing cells.
Congratulations to all my team for this milestone and many many thanks to all co-authors, funding bodies
